| Description |
pBR 322 DNA is isolated from E.coli cloning
(dam+, dcm+) and purified using alkalin method by Phenol/Chlorofom
Extraction. pBR 322 molecule is a double-Stranded circle,
4361 pairs in length.
pBR 322 is one of the most commonly used E.coli cloning
vectors. pBR322 contains: the replicon rep responsible
for the replication of plasmid (sourse -plasmid pMB1 );
the rop gene, coding for the Rop protein, which promotes
conversion of the unstable RNAI - RNAII complex to a stable
complex and serves to decrease copy number (source- plasmid
pMB1); the bla gene, coding for ß- lactamase, that
confers resistance to ampicillin (source- transposonTn3);
the tet gene, encoding tetracycline resistance protein
(source- plasmid pSC101).
pBR 322 can be amplified using chloramphenicol. |
