The InsT/Aclone™ PCR Product
Cloning Kit is a convenient system for direct one-step
cloning of PCR-amplified DNA fragments. Many thermo-stable
DNA polymerase lacking 3'—5' exonuclease activity
"proofreading" possess deoxynucleotidyI transferase
(TdT) and primer extension activity. This frequently results
in the addition of extra deoxyadenosines at 3'-ends of
amplified DNA molecules. These molecules can be ligated
directly to a vector containing compatible single dT-nucleotide
overhangs.
